The A59 at Kex Gill will close overnight this month as work continues to connect the new road.
North Yorkshire Council said the work marks an important milestone towards fully opening the route.
The A59 will be closed between Bolton Abbey roundabout and Menwith Hill crossroads from 7pm to 6am each night from Monday 9th to Friday 13th November.
A second period of overnight closures, covering the same stretch and hours, is planned from Friday 20th to Friday 27th November.
Signed diversions will take drivers via Ilkley and Otley.

Church Hill is due to reopen on Friday 6th November. However, its junction with the A59, along with the Hall Lane junction, will close during the day from Monday 16th to Thursday 19th November for final surfacing.
The A59 itself will remain open to vehicles during that daytime work.
A North Yorkshire Council spokesperson said:
"This is an important milestone in completing the remaining connections and fully opening the road.
"The programme is weather-dependent and dates may need to change at short notice if conditions are unsuitable."
Local access will be maintained where possible during the first period of overnight closures, although routes to individual properties will change as work moves between locations. Affected residents have been sent further details.
Drivers are being asked to allow extra time for journeys and follow the signed diversions.
